![]() ![]() I don't know if this is a state-by-state difference in the punishment or if there is just a massive communication failure from the top-down (which wouldn't be surprising), but my fellow cashiers and I were definitely not aware of such strict punishments. And, if you were wrong on a small winning ticket or two, I would have expected a brief discussion and minor reprimand. I never heard of such a rule with sight validating in fact, my best guess before your post was that it didn't even matter if you sight validated, as long as you got it right. With rules like selling alcohol to a person under 21 years of age, every cashier knew that if you were caught, you would lose your job immediately. That is very interesting, and it probably addresses the root of the issue: communication. TL DR - Clerks pay out a ticket they THINK is a winner because they trust themselves and the lottery machine is always down. I've worked many different jobs at many different levels, and low-level retail in a busy store is easily the most high-pressure of the bunch. However, the reality is that in an environment that employs (typically) people making minimum wage with a high school level education, mistakes and variations from store policy are going to happen. Obviously, this isn't the best practice, and I never saw this happen with a winning ticket of over $5. However, if you work in a store that heavily emphasizes customer service, the customer is a regular customer, you have a long line, and/or you simply don't feel like getting into an argument at that point in time, the pressure is certainly on to just give them their $3 based on your own analysis of the ticket. The official policy is, of course, to tell the customer with a winning ticket that you are sorry that you cannot cash the ticket at that point in time and to come back at some random point in the future when, hopefully, the machine is working. I will never understand why they don't just scan the damn things.Īs a former clerk who sold lottery tickets, I would guess that the reason why "sight" payments is an issue is because the lottery machines (like any other machine in your average convenience store) are down ALL THE DAMN TIME. I will never understand why they don't just scan the damn things.Ĭlerks should never pay out based on a ticket they THINK is a winner. Clerks should never pay out based on a ticket they THINK is a winner. We discourage the hell out of that behavior. ![]() That's a quick path to prison, too.Īnd then, of course, we get idiots who do everything they can to make losing tickets look like winners hoping to get an idiot convenience store clerk to "sight validate" the ticket instead of scanning it in the system to see if it's a win. I've also seen several cases where a retailer is mass producing draw game wagers and re-selling them overseas on the Internet for huge mark-up - sometimes as much as 1000% of face value. What they do is present retailers who players have complained about with "marked" tickets and then have them arrested when the retailer comes in to claim the prize. A few states have even gone so far as to set up an undercover team that specializes in catching these people. You'd be shocked at how often retailers steal tickets from players by telling them that their ticket isn't a winner. ![]() Performing the quantification of Western blots using image software is an essential technique for molecular biologists. Understanding the quantification plot is crucial for analyzing the impact of treatments or experimental conditions on protein expression. A decrease in the quantification plot suggests a reduction in protein expression, while an increase may indicate upregulation. By comparing the normalized ratios of the protein of interest between the control and treatment groups, we can determine whether the expression levels have been Altered. The quantification plot provides valuable insights into the Western blot results. The control group will serve as a reference, and any changes in the treatment group can be easily observed. The graph will demonstrate the relative expression levels of the protein of interest in the control and treatment groups. With the calculated ratios and normalized values, we can plot a graph to Visualize the quantification results. By normalizing, we can compare the relative expression levels of the protein of interest between different samples or treatment groups. This normalization is essential to account for any variations in sample loading and ensure accurate quantification. To normalize the protein of interest, we divide its ratio by the ratio of the housekeeping proteins. Normalizing the Protein of Interest to Housekeeping Proteins Calculation of ratios should be performed for both the housekeeping genes and the protein of interest in both the control and treatment groups. ![]() By dividing the intensity values of the bands of interest by the control value, we can obtain ratios that represent the relative expression levels. In this case, the control value is derived from the housekeeping or actin bands. This involves dividing the intensity values of the bands by a control value. The next step in quantifying Western blots is calculating the ratios. Once the selection is made, we can proceed to the analysis phase. It is crucial to ensure that the selection includes all the bands of interest and that there is enough room underneath for further analysis. The selection should encompass the bands while avoiding unnecessary areas. This is done by using the selection tool, often represented by a rectangle icon. Once the image has been edited to perfection, we can proceed to select the bands of interest for analysis. It may require some trial and error to find the optimal settings. Using the "Adjust" option in the "Image" menu, we can navigate to "Brightness and Contrast" and make adjustments until the background is no longer visible. This can be done by adjusting the brightness and contrast of the image. In addition to aligning the bands, it is often necessary to remove the background from the image to obtain clear curves for analysis. ![]() ![]() Experimenting with different rotation angles will help achieve the desired alignment. If the bands are still slightly tilted, we can use the "Transform" option in the "Image" menu to rotate the image by a specific degree. First, we can invert the image by going to the "Edit" menu and selecting "Invert." This will ensure that the bands appear in a straight line. To correct any alignment issues with the bands, we can use the editing functions of the image software. Once the image is uploaded, it may appear with some imperfections, such as bands that are not aligned or a background that needs to be removed.Įditing the Image: Inverting and Rotating There are two ways to do this - either by going to "File" and selecting "Open" or simply dragging the image into the software. To begin the quantification process, the first step is to upload the image into the image software. Image software allows for accurate and efficient quantification of Western blots by measuring the intensity of the bands. Quantifying the intensity of the bands on a Western blot can provide valuable information about protein expression levels. Western blotting is a widely used technique in molecular biology to detect and analyze specific proteins in a biological sample. In this article, we will discuss how to perform the quantification of Western blots using image software. Solitaire is used in North America while Patience is usually used in the UK and Europe. No, these terms typically refer to the same kind of single-player card games. Is there any difference between Solitaire and Patience? Of course, it’s not always possible to win if you’re unlucky, but a player who knows the right moves at the right time will be able to pull through an otherwise tricky situation. While the game itself does rely on luck and chance a lot, it’s what you do with the draw that matters. Yes, there is a great deal of skill involved in Solitaire. Once you’ve revealed all the cards on the tableau, it’s just a matter of time until you’ve won! After that, it’s about getting better, which is why we provide a timer to see your best time. Otherwise, build your stacks to reveal more cards on the tableau. As you’re going, place any card in the foundations as you can. You’ll be repeating these processes for the rest of the game. Plus, finding Aces in there means you can start building foundations from the tableau or from the stock as well! Remember that once you finish revealing the stock, the waste is placed back into the stock in the exact same order. The stock is a good place to fish for specific cards since you’ll know exactly what’s there. Once you’ve done what you can on the tableau, you can go through the stock. If you can find some Aces this way, that’s great! If you start filling up stacks on top of those face-down cards, it’s going to be harder to reveal them. Beginners will start going through the stock immediately, but these cards aren’t going anywhere. With those rules understood, the best strategy to begin is to reveal as many face-down cards on the tableau as you can. Also, you can only begin a new stack on the tableau with a King, and the waste pile must maintain the same order that you drew them. On the tableau, you must place cards in descending order from King to Ace, and they must alternate colors. The objective of 1 Card Solitaire is to fill your foundations in ascending order from Ace to King by suit. And remember: once you finish your foundations, you win! ![]() Now, you can begin playing! As you can see, the game is all about arranging cards in order. In this version of Solitaire, 1 Card Solitaire, you draw cards from the stock one at a time. When the stock runs out, the waste is brought back into it in the same order. The discard pile next to it is called the waste, where unused stock cards are kept in order of draw. The stack in the upper left is called the stock, which is a pile you can draw from to reveal new cards and add them to the tableau if you’re able. Completing your foundations means you’ve won the game! You must have the appropriate ace down before building on your foundations. The objective of the game is to arrange the cards from the tableau into the four suits-clubs, diamonds, spades, and hearts-in ascending order starting with the ace. When there’s an empty spot, only a king can fill it, so keep that in mind! Revealing all the cards on the tableau is an important strategy of the game. The face-down cards are revealed when there are no face-up cards in front of them. You may arrange cards in descending order on the tableau, switching between suits.įor example, here, you’d be able to move the red queen on top of the black king, but you wouldn’t be able to put the red 5 on top of the red 6. As you can see, there are seven stacks of cards, each with one more face-down card than the next. The cards on the board are called the tableau. In the settings, you can turn autoplay on and off, turn sound on and off, view your game statistics, and view the instructions of the game (just in case the following explanation isn’t enough). One of the main questions students have is what the purpose of an annotation is. The annotation gives information about the relevance and quality of the sources you cited through a 150-250 word description or interpretation of the source.
